JBI-2174 is an orally bioavailable, brain-penetrant small molecule inhibitor of the PD-1/PD-L1 pathway, developed by Jubilant Therapeutics. Unlike monoclonal antibodies that target this pathway, JBI-2174 is designed to cross the blood-brain barrier, making it a potential therapeutic for glioblastoma (GBM) and brain metastases. The compound works by inducing the dimerization of PD-L1, which prevents its interaction with PD-1 and restores T-cell mediated anti-tumor immune responses. In preclinical studies, it has demonstrated significant survival benefits in orthotopic glioma models and comparable efficacy to established anti-PD-L1 antibodies in syngeneic tumor models.
