JBI128 is a first-in-class, dual inhibitor of lysine-specific demethylase 1 (LSD1) and histone deacetylase (HDAC), specifically targeting HDAC1 and HDAC2. Developed by Jubilant Biosys, it acts as an epigenetic modulator that disrupts the CoREST complex, leading to increased expression of differentiation markers (e.g., GFI1b, CD11b) and histone acetylation. Preclinical studies have demonstrated its efficacy across various malignancies, including acute myeloid leukemia (AML), small cell lung cancer (SCLC), and non-small cell lung cancer (NSCLC). Beyond its direct anti-proliferative effects, JBI128 exhibits immune-modulatory properties, showing synergistic activity when combined with immune checkpoint inhibitors like anti-PD-L1 antibodies.
