Overview

JBNm (Janus Base Nanomatrix) is an injectable, cell-free biomaterial scaffold designed for cartilage regeneration and tissue engineering. Developed by researchers at the University of Connecticut and commercialized by Eascra Biotech, it consists of DNA-inspired Janus base nanotubes (JBNts) that self-assemble with extracellular matrix (ECM) proteins (such as matrilins and COMP) and growth factors (such as TGF-β). The scaffold is formed from small molecules (molecular weight below 400 Da) that organize through hydrogen bonding and base stacking at ambient temperature without the need for chemical crosslinkers. JBNm is primarily being developed for the treatment of osteoarthritis and cartilage repair, where it promotes chondrogenesis while inhibiting undesirable processes like hypertrophy, angiogenesis, and dedifferentiation, thereby maintaining long-term cartilage homeostasis.
