JCAR015 is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting the CD19 antigen, originally developed by Juno Therapeutics for the treatment of adult patients with relapsed or refractory B-cell acute lymphoblastic leukemia (B-ALL). The therapy utilizes a gamma-retroviral vector to genetically engineer a patient's own T cells to express a CAR consisting of the SJ25C1 murine anti-CD19 single-chain variable fragment (scFv), a CD28 costimulatory domain, and a CD3-zeta signaling domain. In clinical development, JCAR015 demonstrated significant efficacy, including high complete remission rates in early-phase studies. However, the pivotal Phase 2 ROCKET trial was placed on clinical hold and subsequently terminated in 2017 following multiple patient deaths caused by severe cerebral edema. Celgene (now a subsidiary of Bristol Myers Squibb) acquired the program through its purchase of Juno Therapeutics, but development was discontinued in favor of other CD19-targeted CAR-T candidates with improved safety profiles, such as liso-cel.
