**JCAR017 + avadomide** is an experimental combination therapy for relapsed/refractory B-cell malignancies, most notably aggressive forms of B-cell non-Hodgkin lymphoma. **JCAR017** (also known as lisocabtagene maraleucel, trade name Breyanzi) is a CD19-directed chimeric antigen receptor (CAR) T cell therapy that uses autologous T cells genetically reprogrammed to target CD19-expressing B cells. It is manufactured with a defined composition and separate dosing for CD4+ and CD8+ CAR T cells, intended to optimize efficacy and reduce adverse effects. **Avadomide** (CC-122) is an oral small molecule with immunomodulatory and antiproliferative properties, acting mainly as a cereblon E3 ligase modulator to induce degradation of transcription factors such as Ikaros and Aiolos. The combination is designed to enhance anti-tumor efficacy, leveraging the direct cytotoxic function of JCAR017 and the immune-stimulating, anti-lymphoma effects of avadomide. This combination has been studied in phase 1/2 clinical trials for relapsed/refractory B-cell malignancies.
