Overview

A combination therapy consisting of JCAR017 (lisocabtagene maraleucel), a CD19-directed chimeric antigen receptor (CAR) T-cell therapy, and durvalumab, a programmed death-ligand 1 (PD-L1) blocking monoclonal antibody. JCAR017 is an autologous cell therapy that reprograms patient T-cells to attack CD19-expressing B cells and is used mainly in relapsed or refractory B-cell non-Hodgkin lymphoma. Durvalumab is an immune checkpoint inhibitor that prevents PD-L1 from binding to PD-1 on T-cells, thus enhancing the body’s immune response against cancer. This combination was investigated in clinical trials for relapsed/refractory B-cell malignancies, aiming to enhance anti-tumor immune responses through dual targeting of tumor cells and immune regulatory mechanisms[1][2][3][4][7].
