Overview

JCXH-211 is a first-in-class, lipid nanoparticle (LNP)-encapsulated self-replicating ribonucleic acid (srRNA) therapy encoding engineered human interleukin-12 (IL-12). Developed using Immorna’s proprietary srRNA technology, JCXH-211 is designed for intratumoral or intravenous administration to treat advanced solid tumors. The drug works by enabling prolonged and selective expression of IL-12 in tumor tissues, which modulates the tumor microenvironment and stimulates potent anti-tumor immune responses. Mechanistically, it combines the direct anti-tumor effects of IL-12 with the immune-stimulating properties of srRNA to enhance T cell and NK cell infiltration into tumors while reducing immunosuppressive myeloid-derived suppressor cells. Preclinical studies have shown that JCXH-211 can eradicate established tumors and induce protective immune memory without significant systemic toxicity. Early clinical trials have demonstrated good safety, tolerability, evidence of tumor shrinkage—including abscopal effects—and robust biological activity in heavily pretreated patients with various advanced cancers[1][2][3][5][6][8].
