Overview

JDQ443 + TNO155 is a combination therapy being investigated for the treatment of solid tumors with KRAS G12C mutations. This combination pairs JDQ443, a KRAS G12C inhibitor, with TNO155, a SHP2 inhibitor, to enhance antitumor activity through complementary mechanisms of action. The combination has shown promising results in clinical trials, particularly in patients with non-small cell lung cancer (NSCLC) and other KRAS G12C-mutated solid tumors. JDQ443 is a selective KRAS G12C inhibitor that targets the mutated form of the KRAS protein, which is commonly found in certain cancers, particularly NSCLC and colorectal cancer. TNO155 is a SHP2 inhibitor that blocks SHP2, a protein tyrosine phosphatase involved in multiple signaling pathways. The combination works through two complementary mechanisms: 1. Enhanced target engagement: TNO155 helps shift KRAS to an inactive state, allowing JDQ443 to bind more effectively. 2. Inhibition of pathway reactivation: SHP2 inhibition suppresses feedback activation of wild-type KRAS, NRAS, and HRAS after KRAS G12C inhibition. Preclinical studies demonstrated that this combination improves single-agent activity of JDQ443 in cancer models, with efficacy maintained even at lower doses of either drug. The development of this combination therapy represents an important advancement in targeted therapy for KRAS G12C-mutated cancers, which have historically been difficult to treat effectively with single-agent approaches.
