Overview

JDQ443 + TNO155 + tislelizumab is an investigational combination therapy for **KRAS G12C-mutated solid tumors**. JDQ443 is a small molecule **KRAS G12C inhibitor** that covalently and irreversibly inhibits KRAS G12C mutant protein. TNO155 is a small molecule **SHP2 inhibitor** (targeting the protein tyrosine phosphatase non-receptor type 11, encoded by PTPN11) that disrupts SHP2-mediated RAS/MAPK signaling. Tislelizumab is a **monoclonal antibody** that binds **programmed cell death protein 1 (PD-1)**, blocking its interaction with PD-L1/PD-L2 and thus enhancing T-cell–mediated immune responses. The combination aims to deeply inhibit oncogenic RAS signaling, reduce escape via upstream signaling (SHP2), and enhance antitumor immunity (PD-1 inhibition). It is currently under investigation for efficacy and safety in advanced solid tumors with KRAS G12C mutations, with ongoing enrollment in clinical trials[1][2][4][7].
