JDTic is a synthetic small molecule that acts as a highly selective, long-acting antagonist of the κ-opioid receptor (KOR). It is structurally classified as a 4-phenylpiperidine derivative and is distinct from other KOR antagonists. JDTic exhibits high selectivity for KOR over other opioid receptors such as μ-opioid (MOR) and δ-opioid (DOR) receptors, though some studies report limited selectivity over MOR. Its long duration of action results from altered activity of c-Jun N-terminal kinases rather than irreversible binding. Preclinical studies suggest potential therapeutic applications in depression, anxiety disorders, addiction (including cocaine and morphine), stress-related disorders, psychosis, and obesity. JDTic has been used extensively in research to elucidate the structure of the human KOR[1][3][4][7].
