JEL3108 is a novel, synthetic peptide drug candidate developed by Carocell Bio for the treatment of inflammatory skin conditions, primarily eczema (atopic dermatitis) and burns. The peptide was originally discovered by AstraZeneca and is now being advanced by Carocell Bio. It acts as a highly selective allosteric inhibitor of mitogen-activated protein kinase p38-alpha (MAPK p38α), binding at an allosteric site distinct from traditional small molecule inhibitors. This mechanism allows it to reduce the release of pro-inflammatory cytokines such as TNF-alpha, IL-1beta, and IL-6 in cellular models and human biopsies from patients with eczema. The drug is formulated for topical application using advanced nanoparticle delivery systems to enhance cell penetration and efficacy. Preclinical studies have demonstrated that JEL3108 can significantly improve skin architecture in models of eczema, outperforming comparative steroids in some measures, with potential additional benefits including scar prevention after burns or surgery and delaying skin aging[1][3][6][7][8].
