Overview

JEN-101 is a first-in-class, preclinical immuno-oncology agent designed for veterinary use, specifically in dogs. It consists of fully canine interleukin-12 (IL-12) complexed with aluminum hydroxide. This formulation anchors IL-12 within the tumor microenvironment by binding to collagen, resulting in prolonged local retention and reduced systemic exposure. The primary mechanism is to stimulate a localized immune response against cancer cells while minimizing the severe autoimmune toxicity often associated with systemic cytokine therapies. Preclinical studies and early phase I trials in dogs with advanced melanoma have shown that intratumoral injection of JEN-101 is well tolerated and elicits potent biological activity, including increased infiltration of T cells into tumors and evidence of anti-tumor effects both at treated sites and distant lesions[1][2][3].
