JFAN-1001 is an orally bioavailable, small molecule tyrosine kinase inhibitor (TKI) that targets the epidermal growth factor receptor (EGFR). Developed by Zhejiang Jianfeng Yien Biotechnology, it is primarily indicated for the treatment of advanced non-small cell lung cancer (NSCLC) harboring EGFR mutations, particularly the T790M resistance mutation. JFAN-1001 functions by selectively binding to the ATP-binding site of the mutated EGFR kinase domain, thereby inhibiting downstream signaling pathways such as PI3K/Akt and MAPK that drive tumor cell proliferation and survival. By maintaining high selectivity for mutated EGFR over the wild-type receptor, the drug aims to provide robust clinical efficacy while minimizing common EGFR-related toxicities like skin rash and diarrhea.
