Overview

JHU12-TCR2 is an adoptive T-cell receptor (TCR) engineered T-cell therapy being developed for the treatment of fibrolamellar carcinoma (FLC). It consists of CD4+ T cells engineered to express a specific TCR (JHU12-TCR2) that recognizes the DNAJ-PKAc chimeric oncoprotein, which is the hallmark driver of FLC. This TCR is restricted by the HLA-DRB3*01:01 allele. JHU12-TCR2 was identified from a patient who achieved a near-complete response to a DNAJ-PKAc peptide vaccine and demonstrated superior cytokine production and tumor cell killing in preclinical studies compared to other identified TCRs. The therapy promotes a Th1/Th17-biased differentiation program in CD4+ T cells, which is associated with effective anti-tumor immunity.
