Jiaobentongamine (also known as Giaobentongamine or ZW-001) is a novel, small molecule cardiac myosin activator developed by Guangzhou Zhongwei Biotechnology for the treatment of chronic heart failure. It functions by directly binding to the motor domain of cardiac myosin, which increases the rate of phosphate release and promotes the transition of myosin heads from a weakly bound state to a strongly bound, force-generating state during systole. This mechanism enhances myocardial contractility and improves stroke volume without increasing intracellular calcium concentrations or myocardial oxygen consumption, thereby avoiding the pro-arrhythmic effects and increased metabolic demand associated with traditional inotropic agents like beta-agonists or phosphodiesterase inhibitors. Jiaobentongamine is currently undergoing early-phase clinical evaluation in China.
