JK-1201I is a long-acting polymer-drug conjugate developed by JenKem Technology, consisting of the topoisomerase I inhibitor irinotecan conjugated to a polyethylene glycol (PEG) carrier. This PEGylation is designed to extend the half-life of the active metabolite SN-38, providing sustained therapeutic levels and potentially improving tumor penetration compared to conventional irinotecan. JK-1201I is primarily being evaluated for the treatment of glioblastoma multiforme (GBM), where it is administered intravenously in combination with adjuvant temozolomide following surgery and radio-chemotherapy. In addition to GBM, the drug is being investigated for other oncology indications, including small cell lung cancer, colorectal cancer, and triple-negative breast cancer with brain metastases, with some indications reaching Phase 3 development.
