JK06 is a first-in-class, quadrivalent, biparatopic antibody-drug conjugate (ADC) developed by Salubris Biotherapeutics. It selectively targets the oncofetal protein 5T4 (trophoblast glycoprotein), which is overexpressed in a wide range of solid tumors and associated with poor prognosis and aggressive tumor progression. The ADC consists of an antibody that binds two distinct epitopes on 5T4, enhancing receptor clustering and internalization for more effective delivery of its cytotoxic payload—monomethyl auristatin E (MMAE)—directly to cancer cells while sparing healthy tissue. JK06 is being investigated primarily for unresectable locally advanced or metastatic solid tumors, including breast cancer and bone metastases. As of April 2025, it is in Phase 2 clinical trials in Europe.
