JK07 is a recombinant fusion protein drug developed for the treatment of heart failure, including both heart failure with reduced ejection fraction (HFrEF) and preserved ejection fraction (HFpEF), as well as amyotrophic lateral sclerosis (ALS). It consists of a fully human immunoglobulin G1 monoclonal antibody targeting ErbB3 (HER3) fused to an active polypeptide fragment of human neuregulin 1 (NRG-1). The design allows selective stimulation of the ErbB4 pathway while blocking ErbB3 signaling, aiming to maximize cardiac regenerative effects and minimize side effects associated with NRG-1 therapy. Mechanistically, JK07 acts as an agonist at the ERBB4 receptor and blocks ERBB3 signaling. It is administered intravenously and has shown promising improvements in left ventricular ejection fraction in clinical trials for heart failure[2][3][5][7][9][10].
