JK08 is a first-in-class recombinant fusion protein designed as an anti-tumor biologic for the treatment of advanced and metastatic solid tumors. It consists of two functional elements: a fully human monoclonal antibody directed against cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4) and an interleukin 15/interleukin 15 receptor alpha (IL-15/IL-15Rα) complex. The fusion aims to synergistically enhance anti-tumor immune responses by combining CTLA-4 blockade, which depletes regulatory T cells in the tumor microenvironment, with IL-15-mediated stimulation of CD8+ T cells and natural killer (NK) cells. This dual mechanism is intended to relieve immunosuppression while promoting proliferation and activation of immune effector cells, potentially improving efficacy without significantly increasing immunotherapy-related adverse effects. Developed by Salubris Biotherapeutics, JK08 is administered subcutaneously and has shown preliminary clinical benefit with favorable safety in early-phase trials for heavily pre-treated patients with various solid tumors[1][3][9].
