JK1033 is an investigational small molecule inhibitor of autotaxin (ATX) being developed by Tianjin Jikun Pharmaceutical Technology for the treatment of idiopathic pulmonary fibrosis (IPF). Autotaxin is a secreted lysophospholipase D enzyme, also known as ENPP2, that catalyzes the conversion of lysophosphatidylcholine into lysophosphatidic acid (LPA). LPA is a potent bioactive lipid that promotes fibroblast recruitment, myofibroblast differentiation, and collagen deposition through G protein-coupled receptors. By inhibiting the ATX-LPA signaling axis, JK1033 aims to reduce the pathological fibroblast activity and lung tissue remodeling that characterize IPF, potentially slowing the decline in respiratory function.
