JK36-hcAb is a nanobody-based humanized heavy chain antibody targeting CD38, developed by fusing the llama-derived nanobody JK36 to the hinge and Fc regions (CH2, CH3) of human IgG1. It binds an epitope (E3) on CD38 distinct from those recognized by approved therapies like daratumumab. JK36-hcAb is engineered for use against CD38-expressing tumor cells, mediating potent antibody-dependent cellular cytotoxicity (ADCC) with little complement-dependent cytotoxicity (CDC). Preclinical studies show it suppresses tumor growth and prolongs survival in mouse models of hematological malignancies, suggesting therapeutic potential in multiple myeloma and related disorders[1][2][3][5][7].
