Overview

JMJD2C shRNA is a short hairpin RNA (shRNA) designed to target and knock down the expression of the histone demethylase JMJD2C (also known as KDM4C). JMJD2C acts as a coactivator for hypoxia-inducible factor 1 (HIF-1) and is involved in breast cancer progression by demethylating histone H3 at lysine 9 (H3K9), which enhances HIF-1 binding to hypoxia response elements (HREs). This transcriptional activation upregulates genes required for metabolic reprogramming (such as BNIP3, LDHA, PDK1, and SLC2A1) and lung metastasis (such as LOXL2 and L1CAM). Knockdown of JMJD2C using shRNA has been shown to inhibit breast tumor growth and spontaneous lung metastasis in preclinical mouse models, highlighting its potential as a therapeutic target in oncology.
