Overview

JMKX001899 + IN10018 is a combination therapy under clinical investigation for the treatment of advanced or metastatic non-small cell lung cancer (NSCLC) harboring the KRAS G12C mutation. JMKX001899 (also known as Sosimerasib) is an orally available, covalent inhibitor of the oncogenic KRAS G12C mutation, acting as a targeted therapy with antineoplastic effects. IN10018 (also known as Ifebemtinib or BI-853520) is an orally bioavailable, highly potent and selective small molecule inhibitor of focal adhesion kinase (FAK), which targets pathways involved in tumor proliferation, survival, migration, invasion, and microenvironment modulation. The combination represents a strategy to overcome resistance to KRAS G12C inhibition by targeting complementary oncogenic pathways, aiming to increase clinical efficacy in difficult-to-treat NSCLC. Current development is in Phase 1 clinical trials for KRAS G12C-mutated NSCLC.
