JMKX001899 + IN10018 + pemetrexed + carboplatin is a combination therapy regimen being investigated for the treatment of advanced non-small cell lung cancer (NSCLC) with KRAS G12C mutations. The regimen combines two targeted small molecules—JMKX001899, a selective KRAS G12C inhibitor, and IN10018, a focal adhesion kinase (FAK) inhibitor—with a standard chemotherapy doublet of pemetrexed and carboplatin. JMKX001899 works by covalently binding to the cysteine residue of the KRAS G12C mutant protein, locking it in an inactive GDP-bound state. IN10018 targets FAK, a cytoplasmic tyrosine kinase that is often upregulated in tumors and contributes to survival signaling and resistance to targeted therapies. The addition of pemetrexed (an antifolate) and carboplatin (a platinum-based alkylating agent) provides broad cytotoxic activity by inhibiting nucleotide synthesis and inducing DNA cross-linking. This multi-pronged approach is designed to overcome potential resistance to KRAS inhibition and improve clinical outcomes in patients with KRAS-mutated lung cancer.
