JMKX003142 is a small molecule vasopressin V2 receptor (V2R) antagonist developed by Jemincare and its affiliates, including Guangzhou Jiayue Pharmaceutical Technology. It is designed to treat conditions characterized by fluid retention and renal cyst progression, specifically Autosomal Dominant Polycystic Kidney Disease (ADPKD), heart failure-related fluid retention (cardiac edema), and renal edema. By selectively blocking the V2 receptor in the renal collecting ducts, JMKX003142 inhibits the action of arginine vasopressin, leading to the excretion of electrolyte-free water (aquaresis). In the context of ADPKD, this mechanism also reduces intracellular cyclic AMP levels, which is hypothesized to slow the proliferation of tubular cells and the expansion of renal cysts. The candidate is being evaluated in both oral tablet and intravenous formulations across multiple Phase 1 and Phase 2 clinical trials.
