Overview

JNB274 is a preclinical-stage bifunctional fusion protein developed by JN Biosciences for the treatment of allograft rejection and graft-versus-host disease (GVHD). The molecule consists of an anti-CD122 (IL-2/IL-15 receptor beta subunit) monoclonal antibody fused to the extracellular domain of CTLA4-Ig. This dual-targeting approach is designed to provide synergistic immunosuppression by modulating CD122-expressing effector T cells and natural killer (NK) cells while simultaneously blocking the CD28 costimulatory pathway through the binding of CD80 and CD86 on antigen-presenting cells. By inhibiting both IL-15-mediated survival signals and CD28-mediated activation signals, JNB274 aims to prevent immune-mediated rejection and systemic inflammatory responses in transplant settings.
