JNC-1043 is a first-in-class small molecule inhibitor of importin β (KPNB1) being developed by J&C Sciences for the treatment of triple-negative breast cancer (TNBC), colon cancer, and other advanced malignancies. Importin β is a nuclear transport receptor responsible for the translocation of various proteins, including oncogenic transcription factors, from the cytoplasm to the nucleus. By inhibiting this transport mechanism, JNC-1043 demonstrates potent anti-tumor activity across a range of cancer cell lines and in vivo xenograft models. Furthermore, the compound has shown potential as a radio-sensitizer by blocking radiation-induced cancer cell invasion. Preclinical evaluations in rats and monkeys have indicated a favorable pharmacokinetic profile and safety margin, supporting its advancement as a clinical candidate for oral administration.
