Overview

JNJ-10450232 (also known as NTM-006) is a novel non-opioid, non-NSAID oral analgesic with structural similarity to acetaminophen. It acts as an adenosine A3 receptor modulator and has demonstrated analgesic and antipyretic effects in both preclinical models and human studies. Unlike acetaminophen, it shows no evidence of hepatotoxicity at supratherapeutic doses in preclinical species. The drug is being developed for the treatment of moderate-to-severe acute pain, including postoperative pain. Clinical trials have shown that its onset of action may be slower than acetaminophen but with a longer duration of effect up to 24 hours after administration. The most common adverse event observed was increased bilirubin due to UGT1A1 inhibition; however, no serious adverse events or significant hepatic toxicity were reported[1][3][4][5][6][7].
