JNJ-26070109 is a small molecule antagonist of the cholecystokinin-2 (CCK2) receptor, also known as the CCK-B receptor. Developed by Janssen-Cilag, it was primarily investigated for the treatment of gastrointestinal disorders, including heartburn and abdominal pain. The CCK2 receptor is a G protein-coupled receptor found in the gastric mucosa, where it regulates gastric acid secretion by stimulating histamine release from enterochromaffin-like (ECL) cells. By antagonizing this receptor, JNJ-26070109 was designed to inhibit acid production and potentially address visceral pain. The compound underwent Phase 1 clinical evaluation, including single and multiple ascending dose studies in healthy volunteers to assess its safety, tolerability, and pharmacokinetics, but development appears to have been discontinued.
