Overview

JNJ-27301937 (PEG-CSF1) is a pegylated form of recombinant human colony-stimulating factor 1 (rhCSF-1), also known as macrophage colony-stimulating factor (M-CSF). Developed by Janssen Research & Development, it acts as an agonist at the colony-stimulating factor 1 receptor (CSF1R), which is primarily expressed on cells of the mononuclear phagocyte lineage, including monocytes, macrophages, and osteoclasts. The attachment of a polyethylene glycol (PEG) moiety to the CSF-1 protein significantly extends its circulating half-life and reduces its clearance compared to the native cytokine. JNJ-27301937 was investigated in Phase 1 clinical trials for its ability to stimulate the production and activation of monocytes and macrophages, with potential applications in bone marrow recovery or immunomodulation. Clinical studies were conducted in healthy volunteers and patients with rheumatoid arthritis, but development has since been discontinued.
