JNJ-40929837 is a selective, orally active small molecule inhibitor of leukotriene A4 hydrolase (LTA4H), the enzyme responsible for converting leukotriene A4 to leukotriene B4 (LTB4). LTB4 is a potent chemotactic mediator implicated in the pathogenesis of asthma and other inflammatory conditions. By inhibiting LTA4H, JNJ-40929837 reduces the production of LTB4, thereby targeting inflammation associated with asthma. The drug was developed by Johnson & Johnson and its subsidiary Janssen Research & Development as an anti-inflammatory and antiasthmatic agent. Clinical studies demonstrated that while JNJ-40929837 effectively inhibited LTB4 production in blood and sputum, it did not significantly impact clinical endpoints such as forced expiratory volume (FEV1) in allergen challenge models compared to placebo[2][3][5]. Development for asthma was discontinued after phase II trials[3].
