JNJ-48816274 is a highly selective orexin-2 receptor antagonist (2-SORA) developed as an oral treatment for insomnia. By selectively blocking the orexin-2 receptor, it promotes sleep initiation and maintenance, increasing total sleep time, REM sleep duration, and sleep efficiency while reducing latency to persistent and REM sleep. Clinical studies in healthy volunteers have shown that JNJ-48816274 improves both objective and subjective measures of sleep quality without significant residual effects on cognitive performance. The drug was well tolerated in phase 1 trials. It is being developed by Janssen-Cilag for the potential treatment of insomnia[1][2][3][6].
