Overview

JNJ-55363932 is a small molecule positive allosteric modulator (PAM) of the metabotropic glutamate receptor 2 (mGluR2), developed by Janssen Research & Development. The compound was designed to treat psychiatric disorders by selectively enhancing the activity of mGluR2, a G protein-coupled receptor that modulates glutamatergic neurotransmission. By acting as a PAM, JNJ-55363932 increases the receptor's affinity for its endogenous ligand, glutamate, thereby reducing excessive glutamate release in brain regions like the prefrontal cortex. This mechanism was hypothesized to provide therapeutic benefits in conditions such as schizophrenia and anxiety disorders. Clinical evaluation included a Phase 1 single ascending dose study in healthy volunteers to assess safety, tolerability, and pharmacokinetics, but development appears to have stalled or been discontinued following early-phase testing.
