JNJ-61178104 is a novel human bispecific monoclonal antibody designed to simultaneously target and neutralize both tumor necrosis factor (TNF) and interleukin-17A (IL-17A), two cytokines implicated in the pathogenesis of several autoimmune diseases, including rheumatoid arthritis and psoriatic arthritis. The antibody binds with high affinity to both TNF and IL-17A, blocking their interaction with respective receptors, thereby inhibiting downstream inflammatory signaling. Preclinical studies demonstrated that JNJ-61178104 potently neutralizes TNF and IL‑17A-driven responses in vitro and in vivo models, showing greater efficacy than monotherapy with either parental antibody. Developed using Genmab’s DuoBody technology platform by Janssen Research & Development (a Johnson & Johnson company), the drug advanced to phase I clinical trials for autoimmune disorders but was discontinued before further development[1][2][5].
