JNJ-64530440 is an investigational small molecule drug developed as a capsid assembly modulator for the treatment of chronic hepatitis B virus (HBV) infection. It acts through dual mechanisms of action by inducing the formation of morphologically normal but empty, non-functional HBV capsids (CAM-N), thereby blocking HBV replication as its primary mechanism. Additionally, it inhibits de novo covalently closed circular DNA (cccDNA) formation during early infection as a secondary mechanism. Clinical studies have shown that JNJ-64530440 is well-tolerated and achieves significant reductions in HBV DNA and RNA levels in patients with chronic hepatitis B[1][2][4][5][6][7]. It is being developed by Johnson & Johnson.
