JNJ-67571244 is a novel human bispecific antibody designed to bind both the C2 domain of CD33 and CD3. By engaging these targets, it recruits T-cells to induce cytotoxicity against CD33-expressing cells, which are commonly found in acute myeloid leukemia (AML) and high-risk myelodysplastic syndrome (MDS). The antibody features effector function-silencing mutations in its Fc region to reduce off-target immune cell recruitment. Preclinical studies demonstrated that JNJ-67571244 effectively activates T-cells and mediates killing of AML cells regardless of patient SNP genotype status. It was developed as an investigational therapy for relapsed/refractory AML and high-risk MDS but has been discontinued after phase 1 clinical trials[2][5][6][8].
