JNJ-70218902 is a bispecific T-cell redirecting antibody developed using DuoBody technology. It targets both the transmembrane protein with epidermal growth factor-like and two follistatin-like domains 2 (TMEFF2) on tumor cells and the CD3 receptor complex on T cells. By binding to these targets, it redirects T cells to recognize and kill cancer cells expressing TMEFF2. The drug has been investigated primarily for metastatic castration-resistant prostate cancer (mCRPC). In phase I clinical studies, it demonstrated some clinical activity but was limited by dose-limiting toxicities such as cytokine release syndrome (CRS) and immune effector cell-associated neurotoxicity syndrome (ICANS). No recommended phase II dose was established due to safety concerns[2][4][3].
