Overview

JNJ-73763924 is a chemically synthesized, N-acetylgalactosamine–conjugated short-interfering RNA (siRNA) designed as a targeted therapy for chronic hepatitis B virus (HBV) infection. It is a component of the combination therapy JNJ-73763989, which comprises two liver-targeted siRNA triggers (JNJ-73763976 and JNJ-73763924) that silence viral transcripts through RNA interference. The N-acetylgalactosamine conjugation enables selective hepatocyte delivery via the asialoglycoprotein receptor, providing liver specificity. JNJ-73763924 displays a relatively short plasma half-life, absorption rate-limited kinetics, and high affinity for hepatic uptake, with the combination approach designed to maximize reduction of HBV surface antigen. It is being developed by Johnson & Johnson for subcutaneous administration in patients with chronic hepatitis B.
