Overview

JNJ-73763976 is an N-acetylgalactosamine-conjugated short-interfering RNA (siRNA) molecule that serves as one of the two RNA triggers within the combination investigational product JNJ-73763989. It is designed to specifically target hepatitis B virus (HBV) RNA in liver cells, enabling RNA interference with viral transcripts as a potential antiviral therapy for chronic hepatitis B infection. The mechanism involves liver targeting and gene silencing via the RNA interference pathway. Absorption is mediated by asialoglycoprotein receptor–dependent uptake in hepatocytes. Its clinical development was led by Janssen Sciences Ireland UC (Johnson & Johnson) as part of a combination product for hepatitis B, but development is now discontinued[1][3][5][7].
