JNJ-73763989 (also known as JNJ-3989) is a liver-targeted small interfering RNA (siRNA) therapeutic developed by Janssen Research and Development in collaboration with Arrowhead Pharmaceuticals for the treatment of chronic hepatitis B virus (HBV) infection. The drug utilizes RNA interference (RNAi) to target and degrade HBV viral RNAs, specifically through two N-acetylgalactosamine (GalNAc)-conjugated siRNA triggers: JNJ-73763976 (S-trigger) and JNJ-73763924 (X-trigger). These triggers target the hepatitis B surface antigen (HBsAg) and the hepatitis B x protein (HBx) open reading frames, respectively. By degrading pregenomic RNA and all viral transcripts, JNJ-73763989 aims to reduce levels of all HBV viral proteins, potentially enabling the restoration of the host immune response. It is administered via subcutaneous injection and has been evaluated in Phase 2 clinical trials, including the REEF-1 and REEF-2 studies, frequently in combination with capsid assembly modulators and nucleos(t)ide analogues.
