Overview

JNJ-79635322 + pomalidomide is an investigational combination therapy currently in clinical trials for relapsed or refractory multiple myeloma. JNJ-79635322 (also called JNJ-5322) is a next-generation trispecific antibody that simultaneously binds to B-cell maturation antigen (BCMA) and GPRC5D on myeloma cells, and CD3 on T-cells, enhancing T-cell mediated cytotoxicity against heterogeneous myeloma clones. Pomalidomide is an approved immunomodulatory drug (IMiD) with known anti-myeloma, antiangiogenic, and immunomodulatory effects, acting primarily via cereblon E3 ligase modulation and subsequent effects on lymphocytes. The combination aims to improve efficacy and broaden response rates by attacking myeloma cells through multiple mechanisms. JNJ-79635322 is developed by Johnson & Johnson and its pharmaceutical division Janssen. The combination is undergoing Phase 1b clinical evaluation to determine safety, tolerability, and recommended dosing in patients with multiple myeloma who are refractory to prior lines of therapy[1][3][4][5][7].
