JNJ-80035474 (also referred to as hK2-targeting experimental agent 1 in some preclinical contexts) is an experimental antibody-drug conjugate (ADC) developed by Janssen Research & Development for the treatment of prostate cancer. It specifically targets human Kallikrein 2 (hK2), a serine protease that is highly and specifically expressed in prostate tissue and significantly overexpressed in prostate cancer, including metastatic castration-resistant prostate cancer (mCRPC). The agent consists of a humanized monoclonal antibody directed against hK2, conjugated via a cleavable linker to a potent topoisomerase I inhibitor payload. Upon binding to hK2 on the cell surface, the ADC is rapidly internalized, leading to the intracellular release of the payload, which induces DNA damage and subsequent apoptosis. Preclinical studies have demonstrated robust antitumor activity in xenograft models, and the agent is currently being evaluated in Phase 1 clinical trials for patients with mCRPC.
