JNJ-88549968 is a novel, first-in-class bispecific T-cell redirecting antibody developed for the treatment of calreticulin (CALR)-mutated myeloproliferative neoplasms (MPNs), including essential thrombocythemia and primary myelofibrosis. The drug works by simultaneously binding to the mutated form of calreticulin (CALRmut) on cancer cells and the CD3 antigen on T lymphocytes, thereby redirecting and activating T cells to selectively target and destroy CALRmut-expressing malignant cells. This mechanism aims to enhance immune-mediated cytotoxicity against MPN clones while sparing healthy wild-type cells. JNJ-88549968 is administered as a subcutaneous injection and is currently in Phase 1 clinical trials[1][2][3][4][5][6][7][8].
