Overview

JNJ-89495120 is an investigational small molecule antagonist of the P2X7 receptor (P2RX7) being developed by Janssen Research & Development for the treatment of major depressive disorder (MDD). The P2X7 receptor is an ATP-gated ion channel primarily expressed on microglial cells in the central nervous system, where it plays a critical role in the neuroinflammatory response. Activation of this receptor by extracellular ATP triggers the NLRP3 inflammasome, leading to the maturation and release of pro-inflammatory cytokines such as interleukin-1 beta (IL-1β). Since neuroinflammation is increasingly recognized as a contributing factor to the pathophysiology of depression, JNJ-89495120 is designed to alleviate depressive symptoms by modulating these inflammatory pathways. It is currently being evaluated in the Phase 2 Moonlight-1 clinical trial as a monotherapy for adult patients with recurrent MDD.
