JNJ-98768111 is an antibody-drug conjugate (ADC) developed by Janssen Research & Development for the treatment of advanced prostate cancer. It consists of a humanized monoclonal antibody targeting human kallikrein-2 (KLK2) conjugated to a cytotoxic payload, specifically a topoisomerase I inhibitor. KLK2 is a serine protease that is highly and specifically expressed in prostate tissue and significantly overexpressed in prostate cancer cells, including metastatic castration-resistant prostate cancer (mCRPC). By targeting KLK2, JNJ-98768111 is designed to selectively deliver its cytotoxic payload to tumor cells, where it is internalized and released to induce DNA damage and cell death. The agent is currently being evaluated in a Phase 1 clinical trial (NCT05924217) to determine its safety, tolerability, and recommended Phase 2 dose in patients with advanced prostate cancer.
