Overview

JNJ28610244 is a potent and highly selective small molecule agonist of the Histamine H4 receptor (H4R). Developed by Janssen Research & Development, it serves as a critical pharmacological tool for exploring the therapeutic potential of H4R activation in oncology and inflammatory conditions. In preclinical studies, particularly in metastatic melanoma and triple-negative breast cancer models, JNJ28610244 has demonstrated significant anti-tumor activity by inhibiting cell proliferation, inducing senescence, and promoting terminal differentiation (melanogenesis). It also exhibits anti-metastatic properties by reducing the expression of histidine decarboxylase and the activity of matrix metalloproteinase-2 (MMP2). Its high specificity for the H4R subtype distinguishes it from earlier histamine ligands, making it a valuable candidate for investigating H4R-mediated signaling pathways.
