JNK1 ASO is an antisense oligonucleotide (ASO) specifically designed to target and reduce the expression of Jun N-terminal kinase 1 (JNK1) mRNA. By binding to the complementary sequence of JNK1 mRNA, it promotes its degradation or inhibits its translation, thereby decreasing the production and activity of the JNK1 protein. In preclinical studies, administration of JNK1 ASO in obese mouse models led to a significant reduction in JNK1 mRNA and activity in liver and adipose tissues, resulting in lower body weight, reduced fat content, improved insulin sensitivity, increased metabolic rate, decreased plasma cholesterol levels, and improved liver steatosis. The mechanism involves limiting caspase-dependent apoptosis signaling pathways under hypoxic conditions as well as modulating genes involved in lipid metabolism[3][6]. This approach suggests potential clinical benefit for treating obesity and related metabolic disorders by increasing fuel combustion and decreasing lipogenesis[6].
