JO-122 is a novel tropolone derivative, chemically identified as 2-(1,1-dimethyl-1H-benzo[e]indoline-2-yl)-5,6,7-trichloro-1,3-tropolone, currently under investigation for the treatment of high-grade gliomas. Developed by researchers at the National Medical Research Centre for Oncology and South Federal University in Russia, it belongs to a class of tropolone alkaloids noted for their broad pharmacological potential. Preclinical in vitro studies have demonstrated that JO-122 exerts a pronounced cytostatic and cytotoxic effect on primary cultures of grade 4 glioblastoma and anaplastic astrocytoma cells, showing time-dependent reductions in cell viability with IC50 values ranging from approximately 1.1 to 5.4 μmol.
