JOAB-40 is a novel small molecule enaminone derivative belonging to the JOAB series, developed as an anti-inflammatory agent. It was designed by modifying the parental compound E121 to include a piperazino ring. JOAB-40 exerts its therapeutic effects by reducing the levels of pro-inflammatory cytokines, including TNF-α, IL-1β, IL-6, and IL-12, and by inhibiting chemokine signaling through the C-C chemokine receptor type 2 (CCR2). In preclinical studies, JOAB-40 has demonstrated efficacy in reducing the severity of colitis in murine models (DSS-induced) and has shown potential in treating inflammation associated with hemarthrosis. It appears to be more potent than its predecessor E121, particularly in treatment-based approaches for colitis.
