JP11646 is a novel small molecule pan-PIM kinase inhibitor developed to target PIM2, a serine/threonine kinase found to be overexpressed in several types of solid cancers and hematological malignancies. Its mechanism of action includes proteasome-dependent degradation of the PIM2 protein, leading to reduced downstream oncogenic signaling and induction of apoptosis in cancer cells. Preclinical studies demonstrated that JP11646 can significantly suppress tumor growth and cancer cell proliferation both in vitro and in vivo, particularly in models of breast, liver, pancreatic, and non-small cell lung cancer, with minimal observed toxicities at effective doses. JP11646 appears to be more effective than earlier PIM kinase inhibitors by virtue of not only reducing PIM2 kinase activity but also the protein level itself. The drug is under investigation as a potential treatment of multiple solid cancers, but there are no reports of clinical trials or regulatory approval as of 2025.[1][3][4][6][8][10]
